
Enzovera 10× Running Buffer with EDTA (EV-SGR-006) is a 10× concentrated capillary electrophoresis running buffer formulated for ABI Genetic Analyzers. It maintains optimal ionic conditions throughout the CE separation, providing the stable electrolyte environment required for high-resolution fragment separation. The buffer is diluted to 1× working concentration immediately before use. A 500 mL bottle yields 5 L of 1× running buffer.
Buffer composition and function. The 10× formulation contains a buffered electrolyte system at controlled pH with EDTA as a divalent cation chelator. EDTA sequesters magnesium and other divalent cations that would otherwise interact with DNA during electrophoresis, ensuring that fragments migrate purely on the basis of size rather than ion-mediated interactions with the polymer matrix or capillary wall. The buffer maintains pH stability and ionic strength throughout extended CE runs (4–6 hours on long-capillary configurations), preventing the resolution loss and peak broadening that occur with degraded or pH-drifted buffer.
Dilution and use. Dilute 100 mL of 10× concentrate with 900 mL of nuclease-free deionized water to prepare 1 L of 1× working buffer. Fill the cathode and anode buffer reservoirs to the recommended fill lines on the instrument. Replace running buffer every 24–48 hours of instrument use, or as specified by the instrument manufacturer's maintenance schedule.
Compatibility. EV-SGR-006 is compatible with all ABI capillary electrophoresis Genetic Analyzers including the 3100, 3130, 3130xl, 3500, 3500xl, 3730, and 3730xl systems. It is suitable for both Sanger sequencing runs (paired with POP-7, EV-SGR-004) and fragment analysis runs (paired with POP-4, EV-SGR-005).
Recommended applications:
Pack size: 500 mL bottle (10× concentrate). Store at 2–8°C. Dilute 1:10 with nuclease-free deionized water before use. 1× working buffer is stable at 2–8°C for 1 month after dilution.
