VeraDye Terminator v3.1 Cycle Sequencing Kit
Enzovera VeraDye™ Terminator v3.1 Cycle Sequencing Kit (EV-SGR-002) is an improved v2.0 cycle sequencing enzyme mix designed as a drop-in replacement for BigDye® Terminator v3.1. The kit delivers enhanced signal uniformity and reduced dye blob artifacts, producing read lengths exceeding 900 bases on ABI 3730xl capillary electrophoresis instruments. It is engineered for compatibility with GC-rich and homopolymer templates that often present challenges to standard sequencing chemistries.
Cycle sequencing chemistry. The kit contains a thermostable DNA polymerase, four dye-labeled dideoxynucleotide chain terminators (ddA, ddC, ddG, ddT), unlabeled dNTPs, magnesium, and proprietary stabilizers in a single ready-to-use master mix. Each of the four terminators carries a distinct fluorescent label spectrally compatible with ABI 3xxx-series detection optics, allowing all four reactions to be combined in a single tube and read in a single CE injection. The terminator-to-dNTP ratio is balanced to deliver uniform peak heights across the full read range, eliminating the early-read dye blobs and late-read signal dropoff characteristic of less-optimized chemistries.
Performance on difficult templates. VeraDye™ Terminator v3.1 has been optimized to deliver clean reads through GC-rich regions, homopolymer runs, and templates with strong secondary structure that often cause compression and signal dropout with standard chemistries. For exceptionally difficult templates (high-GC content above 70%, long homopolymeric stretches, or strong hairpins), the kit can be supplemented with HP Hairpin Reagent (EV-SGR-010, 7-deaza dATP) to further destabilize template secondary structure.
Instrument compatibility. VeraDye™ Terminator v3.1 is fully compatible with all ABI 3xxx-series capillary electrophoresis instruments including the 3130, 3130xl, 3500, 3500xl, 3730, and 3730xl Genetic Analyzers. Use POP-7 Separation Polymer (EV-SGR-004) on 3730/3730xl instruments and POP-4 Separation Polymer (EV-SGR-005) on 3100/3130/3500 systems. The kit uses standard ABI mobility files and dye sets — no instrument re-calibration is required for switching from BigDye® v3.1.
