Sequencing Product Purification Magnetic Beads

Enzovera Sequencing Product Purification Magnetic Beads (EV-SGR-007) are silica-hydroxyl–surface magnetic beads developed for purification of Sanger sequencing extension products prior to capillary electrophoresis injection. They remove unincorporated dye terminators, salts, and short fragments that would otherwise contaminate the CE injection, producing clean electropherograms with low baseline noise and accurate base calling from the first injectable nucleotide. The product delivers performance equivalent to CleanSEQ (Beckman Coulter) and is compatible with both manual and automated liquid handling platforms.

Binding and cleanup mechanism. The silica-hydroxyl surface chemistry selectively binds long extension products (sequencing fragments >50 bases) in the presence of an alcohol-based binding solution, while unincorporated dye terminators, free dNTPs, magnesium salts, primer fragments, and short failed-extension products remain in solution and are washed away. The bound extension products are then eluted in low-salt elution buffer or directly resuspended in Hi-Di Formamide (EV-SGR-011) ready for CE injection. Total cleanup time is approximately 15 minutes from reaction completion to CE-ready plate.

Performance specifications. Dye removal efficiency exceeds 95% under standard protocol conditions, eliminating the dye blob artifacts that obscure early reads on uncleaned sequencing reactions. Recovery of sequencing extension products is typically >90%, preserving signal intensity for full read length recovery. The beads are supplied as a uniform aqueous suspension and require only brief vortexing before use.

Automation compatibility. EV-SGR-007 is compatible with all major automated liquid handling platforms including Beckman Biomek, Hamilton STAR, Tecan Freedom EVO, and Eppendorf epMotion systems. The bead suspension dispenses cleanly through standard tip geometries and the magnetic separation step uses standard 96- and 384-well plate magnets. For manual workflows, a benchtop 96-well magnet is sufficient. 10,000 reactions per tube supports high-throughput core facility operations with minimal reagent change-out frequency.

Recommended applications:

Pack size: 10,000 reactions per tube. Store at 2–8°C. Do not freeze. Vortex briefly to resuspend before use.
